di restituire la speranza a milioni di persone affetta da Alzheimer. I ricercatori, infatti, avrebbero scoperto un trattamento rivoluzionario in grado di alleviare i sintomi della malattia. Il farmaco, stando a quanto pubblicato sulle pagine della rivista Science Signaling, è stato testato con successo sui topi, e si basa sull’uso della dopamina. La sostanza chimica, preposta alla trasmissione dei segnali nel cervello e in altre aree del sistema nervoso, potrebbe favorire la produzione di neprilisina, un enzima in grado di scomporre le placche beta-amiloidi nel cervello, riducendo così i danni neuronali caratteristici dell'Alzheimer.Nuovi orizzonti terapeutici Guidato dal professor Takaomi Saido, il team di ricerca ha osservato che la dopamina aumenta i livelli di neprilisina nelle cellule cerebrali coltivate in laboratorio, e successivamente nei cervelli dei topi trattati. Questo enzima è cruciale perché scompone le placche beta-amiloidi, accumuli proteici che danneggiano i neuroni e sono strettamente legati alla progressione dell'Alzheimer. Gli esperimenti sui topi hanno mostrato una significativa riduzione delle placche nella corteccia prefrontale dopo otto settimane di trattamento con dopamina, accompagnata da miglioramenti nella memoria.La promessa di L-DOPASebbene la manipolazione genetica per aumentare la neprilisina non sia praticabile negli esseri umani, i ricercatori hanno testato l'uso di L-DOPA, un precursore della dopamina già utilizzato nel trattamento del Parkinson. Nei modelli murini, il trattamento con L-DOPA ha non solo ridotto le placche beta-amiloidi nel cervello, ma ha anche migliorato le prestazioni nei test di memoria. Questo risultato apre la possibilità di utilizzare L-DOPA per contrastare l'Alzheimer, benché siano necessari ulteriori studi per valutarne la sicurezza e l'efficacia negli esseri umani.Sfide e prospettive futureWatamura Naoto, primo autore dello studio, ha sottolineato che, nonostante i risultati promettenti, il trattamento con L-DOPA presenta notevoli effetti collaterali nei pazienti con Parkinson. "Il nostro prossimo passo", spiega Naoto, "è indagare come la dopamina regola la neprilisina nel cervello. Questo potrebbe portare a nuovi approcci preventivi che potrebbero essere avviati nella fase preclinica dell'Alzheimer".
