"Grazie a un lavoro di squadra che ha visto coinvolti la dottoressa Laura Giunti (Neuro Oncologia), la dottoressa Anna Maria Buccoliero (Anatomia Patologica), il dottor Iacopo Sardi (Neuro Oncologia) e il dottor Lorenzo Genitori (Neurochirurgia), l’Aou Meyer Irccs è adesso in grado di valutare il profilo di metilazione (metiloma) delle patologie oncologiche che colpiscono, spesso in modo aggressivo, anche i bambini - sottolinea una nota dell'Aou Meyer - Grazie a un ingente investimento tecnologico e alla competenza dei suoi operatori, l’ospedale Meyer si allinea così ai migliori standard mondiali per la diagnosi e conseguentemente per la terapia di questi tumori rari e ancora poco conosciuti. La metodica introdotta richiede infatti personale altamente specializzato in ambito biomolecolare e bioinformatico e l’impiego di sofisticate attrezzature di nuova generazione e di complessi algoritmi informatici". Come funziona. "La metodica rileva la presenza di gruppi metilici in particolari regioni del Dna chiamate isole di metilazione - continua il Meyer - La metilazione di queste regioni genomiche regola l’espressione genica vale a dire lo stato “acceso” o “spento” dei singoli geni. È noto che ogni tumore ha un proprio specifico profilo di metilazione che lo distingue da tutti gli altri tumori, pertanto l’analisi dello stato di metilazione del suo DNA fornisce informazioni utili e in qualche caso indispensabili ai fini diagnostici, prognostici e predittivi. L’iter sperimentale parte dall’estrazione del DNA da un piccolo frammento tumorale asportato chirurgicamente e si conclude con il confronto del profilo di metilazione del tumore in esame con quello di un grande numero di tumori di riferimento al fine di trovare la precisa corrispondenza diagnostica".L’introduzione di questa analisi è particolarmente utile per la diagnosi dei tumori con numerosi sottotipi molecolari la cui precisa classificazione ha importanti ricadute prognostico-terapeutiche o in quei casi di tumori in cui le tradizionali metodiche di diagnostica anatomo-patologica si rivelano non conclusive per una precisa ed accurata definizione diagnostica."Attraverso l’analisi del profilo di metilazione è possibile inoltre identificare aberrazioni cromosomiche che aiutano i medici nella scelta del più efficace e personalizzato trattamento farmacologico post-chirurgico. Non per ultimo, tale metodica renderà possibile riconoscere e classificare nuove entità tumorali che allo stato attuale delle conoscenze non trovano una corrispondenza diagnostica tra i tumori noti", evidenzia il Meyer. "Lo studio del profilo di metilazione non ha solo valenza diagnostica, prognostica e terapeutica ma rappresenta anche un utile strumento per la ricerca scientifica.
